Morphological and cultural properties of a medicinal mushroom, Piptoporus betulinus (Basidiomycetes), on nutrient agar media

Abstract

The article presents results of the research on growth and morphological peculiarities of a valuable medicinal mushroom, Piptoporus betulinus, from the Culture Collection of Mushrooms of M.G. Kholodny Institute of Botany, National Academy of Sciences of Ukraine. Using scanning electron microscopy, the micro-morphological structures specific to this species were studied in order to enable its identification in pure culture. Cultural-morphological studies were performed on four nutrient agar media. For all tested strains, potato-glucose agar medium (PGA) and incubation temperature of 26º C were the most favorable for vegetative growth. Temperature of 36° C is critical. We have studied the presence of eight enzymes characterizing: redox processes (laccase, tyrosinase, peroxidase); metabolism of nitrogen compounds (protease); carbohydrates (amylase, cellulase, β-glucosidase); lipids (lipase). The established cultural  characteristics can be used for verification of P. betulinus cultures.
